| Transfect 200ng of PiggyBac mRNA and 800ng of donor DNA with eGFP sequence into 5×104 HEK293T cells using Lipofectamine™ 2000 (or equivalent). On day 3, 6, 10 and 12, use flow cytometry analysis to evaluate the ratio of eGFP positive cells and eGFP expression intensity. |
