Description

A linker is an amino acid chain that connects two fused proteins, allowing for the appropriate separation of protein functional domains or the maintenance of necessary inter-domain interactions. The most widely known flexible linker is the poly-Glycine-Serine (GS) linker. The GS Linker includes various forms, such as (GS)n, (GGS)n, (GGGS)n, (GGGGS)n, etc. These GS linkers are widely used in connecting bispecific antibodies, antibody fusion proteins, single-chain antibodies, peptides and ADCs, etc.
GenScript’s Anti-GS Linker Affinity Resin FF (Cat. No. L01036) is designed for purification of proteins, antibodies and compounds containing (GnS)m linker (n≥2, m≥2) from commonly used protein expression systems such as bacteria, yeast and mammalian cells. The GS linker fusion protein in cell lysate can specifically bind to the anti-GS linker antibody coupled to the resin. Nonspecific binding reactions are eliminated by stringent washing steps, and the purified target protein is eluted with a high recovery rate. Table 1 lists the main characteristics of Anti-GS Linker Affinity Resin FF.

Resin reuse When properly regenerated and stored according to the instructions, the resin can be reused up to 10 times without significant loss of binding capability.
Elution method Acidic elution (pH 3.0)
Reagent compatibility Compatible with commonly used lysis reagents. For reagents compatible with purifications, see tables 2 and 3.

Anti-GS Linker Affinity Resin FF

The target protein containing (G4S)3 linker was captured by GenScript Anti-GS Linker Affinity Resin FF (Cat. No. L01036). The resin was washed twice with 10 times bed volume of TBS and then eluted with 10 times bed volume of acidic eluent (0.1 M glycine HCl, pH 3.0). Subsequently, the resin was regenerated by 10 times bed volume of regeneration buffer for repeated use. The coefficient of variation (CV) of the amount of eluted target protein was less than 20% for a total of 10 purification processes, indicating that GenScript Anti-GS Linker Affinity Resin FF is capable of multiple repeated use with relatively high consistency when eluted by acidic eluent. »
